narlox (naloxone hydrochloride) Injection, USP

NARLOX® Naloxone is a drug used to counter the effects of overdosing with opioids such as heroin and morphine. Specifically, naloxone is used in opioid overdoses for countering life-threatening depression of the central nervous system and respiratory system. The drug is derived from baine and has an extremely high affinity for µ-opioid receptors in the central nervous system.

NARLOX® Naloxone is a µ-opioid receptor competitive antagonist and the rapid blockade of these receptors often produce rapid onset of withdrawal symptoms. NARLOX® Naloxone is injected usually through intravenous route for fastest action. The drug acts after about two minutes and its effects may last approximately 45 minutes. Naloxone has been shown to reduce rates of fatal overdose with heroin.

NARLOX (Naloxone Hydrochloride)

Naloxone is a an opioid antagonist drug. used to counter the effects of opiate overdose, for example heroin or morphine overdose. Naloxone is specifically used to counteract life-threatening depression of the central nervous system and respiratory system. Naloxone is also experimentally used in the treatment for congenital insensitivity to pain with anhidrosis (CIPA), an extremely rare disorder (1 in 125 million) that renders one unable to feel pain.

NARLOX® Naloxone has an extremely high affinity for μ-opioid receptors in the central nervous system. Naloxone is a μ-opioid receptor competitive antagonist, and its rapid blockade of those receptors often produces rapid onset of withdrawal symptoms. Naloxone also has an antagonist action, though with a lower affinity, at κ- and δ-opioid receptors.

NARLOX® Naloxone is most commonly injected intravenously for fastest action. The drug generally acts within a minute, and its effects may last up to 45 minutes. It can also be administered via intramuscular or subcutaneous injection. Use of a wedge device (nasal atomizer) attached to a syringe to create a mist delivering the drug to the nasal mucosa [2] - may also be utilized, although this solution is more likely utilized outside of a clinical facility.

What is NARLOX™ (naloxone)?
NARLOX™ (naloxone) is an opiate antidote. Opioids include heroin and prescription pain pills like morphine, codeine, oxycodone, methadone and Vicodin. When a person is overdosing on an opioid, breathing can slow down or stop and it can very hard to wake them from this state. NARLOX™ (naloxone) is a prescription medicine that blocks the effects of opioids and reverses an overdose. It cannot be used to get a person high. If given to a person who has not taken opioids, it will not have any effect on him or her, since there is no opioid overdose to reverse. How does NARLOX™ (naloxone) work?
If a person has taken opioids and is then given NARLOX™ (naloxone), the opioids will be knocked out of the opiate receptors in the brain. NARLOX™ (naloxone) can help even if opioids are taken with alcohol or other drugs. After a dose of NARLOX™ (naloxone), the person should begin to breathe more normally and it will become easier to wake them. It is very important to give help to an overdosing person right away. Brain damage can occur within only a few minutes of an opioid overdose as the result of a lack of oxygen to the brain. NARLOX™ (naloxone) gives concerned helpers a window of opportunity to save a life by providing extra time to call 911 and carry out rescue breathing and first aid until emergency medical help arrives.

How is NARLOX™ (naloxone) given to an overdosing person?

NARLOX™ (naloxone) can be given by intramuscular (IM) injection - into the muscle of the arm, thigh or buttocks - or with a nasal spray device (into the nose). In Illinois, NARLOX™ (naloxone) is prescribed for use as an IM injection. Nasal spray use is less common, but some large cities in the U.S. use the nasal spray version and it can be prescribed.

How long does NARLOX™ (naloxone) take to work?

NARLOX™ (naloxone) generally works within about 5 minutes. Repeated doses may be necessary if a person is still showing signs of overdose even after the first dose. Learn how to use naloxone here.
How long does NARLOX™ (naloxone) take to wear off?
NARLOX™ (naloxone) starts to wear off after about 30 minutes and is mostly gone after about 90 minutes. By this time the body has processed enough of the opioids that the overdosing person is unlikely to stop breathing again. In some cases, such as after taking a massive dose or using long-acting opioids like methadone, the patient might need another NARLOX™ (naloxone) dose and longer medical observation. Always watch the person after they receive a NARLOX™ (naloxone) dose for signs of continued overdose.

Who can be prescribed NARLOX™?

According to the Overdose Prevention Act, trained individuals are allowed to possess and administer NARLOX™ (naloxone) to a person having an overdose. Physicians may also train individuals who can then go on to train other individuals (this is called “train the trainer” model). Currently, the main NARLOX™ (naloxone) provider in Illinois is the Chicago Recovery Alliance. But there are a growing number of programs and physicians available throughout the state and more laypeople are being trained in naloxone use all the time. NARLOX® Naloxone has been distributed as part of emergency kits to heroin users, and this has been shown to reduce rates of fatal overdose. Projects of this type are under way in San Francisco, New Mexico, Philadelphia, New York State, Baltimore, Boston, Los Angeles, Milwaukee, and Chicago, with pilot projects started in Scotland in 2006. The combination oxycodone/naloxone is used for the treatment of opioid-induced constipation under the trade name Targin and in the Netherlands under Targinact.

Legal status : The patent for NARLOX® Naloxone has expired and the drug is currently available in various generic forms.
