COMPOSITION :
Allopurinol 100

INDICATIONS
:
* Treatment of hyperuricemia associated with primary gout and
secondary gout ( uric acid lithiasis, nephropathy,
tophi, joint destruction).
* Treatment of hyperuricemia associated with leukaemia, resulting
from radiotherapy, the use of anti-neoplastic agents such as
mercaptopurine or during treatment with diuretics of the thiazide
or similar type.
* Treatment of children with hyperuricemia secondary to malignancy
or to certain rare inborn errors of purine metabolism.
DOSAGE AND ADMINISTRATION :
In gout, an initial daily dose of 100mg is increased by 100mg at
weekly intervals to maximum of 800mg daily in divided doses.
In hyperuricemia associated with leukaemia a suggested initial
dose is 200 mg thrice daily commencing if possible 2 or 3 days
before radiotherapy or the commencement of treatment with anti-neoplastic
agents, and adjusted as requested to a maintenance dose usually of
300 to 400 mg daily. Allopurinol treatment should be discontinued
when the potential for overproduction of uric acid is no longer
present.
For children the suggested initial dose is 8-20 mg per kg body
weight daily. The dose should be titrated against the serum urate/uric
acid and or urinary uric acid levels at appropriate intervals.
Fluid intake should be sufficient to maintain daily urinary volume
above 2 litres.
Dosage must be reduced in patients with renal impairment in
proportion to the reduction in glomerular filtration.
CONTRAINDICATIONS :
* Patients who have exhibited serious adverse effects from the
medication
* Known intolerance to Allopurinol
* Severe hepatic or renal disorder
* Children (except those with malignancy)
* As treatment of acute gout
